My sister has a faint recollection of a beloved childhood PC game in which the character wore a yellow helmet and fought Far Side-esque green slugs, in a "space building." ("Maybe not the yellow helmet, but definitely green slugs.") Ring any bells?

Heck yeah Commander Keen! One of the first games by id Software, those who brought us Wolfenstein, Doom, Doom II, The Great Big Disc of Doom Wads, Heretic, Hexen: Further Heretic, Quake, Quake II: The Quakening, Quake III: Son of Quake, Return to Castle Wolfenstein, Wolfenstein: Enemy Territory, Doom III: Flashlight Future Soldier!, Quake IV: Return to Quake, Enemy Territory: Quake Wars, ummm...Wolfenstein again...and, next up, QuakeDoom Territory: Borderlands Fallout. I mean, Brink.
Your sis might have also played the original Duke Nukem platformers, and will possibly squee with a similar level of delight upon being reminded of it.

More specifically, that was episode 4, "Secret of the Oracle". The second series (episodes 4 and 5) is much different from the first.
