I'll be traveling from Baton Rouge, CA to Hattiesburg, MS on either a Friday evening or a Saturday morning, and I'm looking for the best way (most convenient and cost-effective) to get there.

Do you mean Baton Rouge, Louisiana? I can't find a town in California with that name.

Rome2Rio says a bus to New Orleans and then a train to Hattiesburg will take 4-5+ hours and cost up to $125.

Rome2Rio seems to have the wrong schedules. The train from New Orleans leaves at 9:15 AM and Rome2Rio says it leaves at 7 AM. (There is only one train a day. Fortunately the northbound train starts in New Orleans!)

But that's good, because that 7 AM departure would mean you'd need an overnight stopover in New Orleans. It looks like Greyhound has a bus leaving Baton Rouge at 6:00 AM, arriving New Orleans 7:57 AM, and the New Orleans bus station and train station are right next to each other.

Do you need to get back? In that case you do the same thing in reverse. Get on a train in Hattiesburg at 5:53 PM, arriving New Orleans 9:02 PM. Bus to Baton Rouge leaves 11:00 PM, arriving Baton Rouge 12:15 AM - this is FlixBus. Again there is only one train a day from Hattiesburg to New Orleans. The train originates in New York, though. Hopefully it is not running late.

I mean, the best way in terms of time/cost is to rent a car. This is not a region in the country with good public transportation.

Ummm, no. A bus has many stops, but it probably won't be stuck for hours. Passenger trains are notorious for delays.
Bus routes are more reliable.

You have a good point - especially if you're talking about a southbound train which has more opportunity to be running late - but as far as I can tell Hattiesburg doesn't have any intercity bus service.

Yes, rent a car. I-12 to I-59.

Driving from Baton Rouge to Hattiesburg is pretty easy. Any other option will be a real trial, especially in summer. If a rental car is not an option, I would seriously price out a taxi or Uber before considering the bus or train.
