Preston and Steve do the best morning show ever. They are consistently funny without being insulting or derogatory. The put up about 2 hours of great comedy a day for free. Current shows and a select few classic bits are on iTunes, and older shows are archived on prestonandsteverock.com . In fact, last thursday's show (19Mar09) was great. Start there.

I'm sure you'll see this one a lot, but Radiolab!!! It's like This American Life with a science theme.
That being said, This American Life is another winner for me.
The Best of the Chris Moyles Show. He's the breakfast show dj on BBC Radio 1 and it's really hilarious. He has a lot of famous guests.
Hey We're Back is a podcast by Jonathan Katz. It's short and updated unregularly, but it's hilarious.
Wait, Wait Don't Tell Me would probably be my #5. But NPR has a ton of great podcasts.

I also sometimes like Diggnation, but haven't been watching it as of late.

The Ricky Gervais show is the funniest thing I have ever listened to.
Dangerous to listen to while driving as you might find yourself laughing so hard you have taken your eyes off the road. And yes, people in traffic will stare.

Laughing just thinking about it...battle dolphins, bees having heart attacks, the giant diary...
