What 80's movie is this quote from and what exactly is the right quote? Its something like "I've got a girlfriend, we met at Niagara Falls. You wouldn't know her."

The "girlfriend in/from Canada" quote is from BClub.
But when refenced, it's not necessarily a BClub reference.
It references the more general phenomenon of a fictional girlfriend/boyfriend for those who lie about not being virgins and can't produce a sex partner.

Summer Camp was often used before Canada:
Lori: You're such a virgin!
Lynn: Am not!
Lori: Oh yeah, who did you have sex with? When?
Lynn: um... it was last summer, with this boy at camp; you don't know him.

The Broadway musical "Avenue Q" also references this meme, with a closeted gay character singing "oh I wish you could meet my girlfriend / my girlfriend who lives in Canada".

I think the Weird Science one is definitely a references since both men with imaginary girlfriends are played by the same actor.
