What is one of the hardest problems in your field of work or study? Why is it so hard?

My nomination for Biomedical Science:
Protein folding - understanding how a string of amino acids takes on it's 3-dimensional shape is an insanely difficult problem involving some serious molecular biophysics. If we could better understand this phenomenon, we could understand why proteins sometimes misfold (and lead to a whole slew of disorders and diseases).
