I was looking at exactly the same thing this afternoon. Check out all the AskMeFi questions tagged with Prague. That got me going - also getting a Lonely Planet guide is a good idea.

I already asked this.

From the responses I got there:

We did the funicular railway/view of the city thing: recommended. There's also a fun mirror maze up there.

Sedlec Ossury - excellent.

Sorry for self-link, but this is our account of our week in Prague in July.

I would call the Old Jewish Cemetery a must see.

You'll probably see a pile on here about the Sedlec Ossuary. But that only works if you have the mobility to get out there.

Off the top of my head, the most memorable part of my brief visit to Prague was the big castle/cathedral across the river, and the little alley of shops below it.

Getting out there is just a train ride, and a bus (or walk). Its definitely worth it.

The Alphonse Mucha Museum was wonderful. If you like art and design, it's worth a few hours. Also, Praque is a great place to see cubist architecture, and a great place to wander around in general. Just mark a few things you want to see on your map each day and then slowly make your way around to see them.

Also, what Orange Goblin said; it's *very* quick and easy to get to Sedlec/Kutna Hora from Prague by public transport. It's a very cool day trip, even if lots of tourists do it.

I second just about everything everyone has said: The Castle, the Jewish Cemetery, the Mucha Museum. It's probably going to be really cold while you're there, but try to do as much walking as you can. And buy a public transportation pass for the time you're there. It's a lot easier than buying tickets for every ride, and you'll feel more free to jump on a tram at a moment's notice. The Ossuary/Sedlec is a good trip to make if you'd like to get out of town. Bring a map with you so you can find your way from the Ossuary into town to see the sights there: St. Barbora's church and the Silver Mine are worth doing while you're there.
