Most businesses tend to use American Express. I don't know why.

AMEX, but if you really like air miles go with Chase Visa

Just from experience dealing with business travelers, AmEx is the most popular for small or mid size businesses. Your employees will also be frustrated once in a while if they find places that don't take AmEx for various reasons. Not particularly common, but if you go with AmEx, it wouldn't hurt to have a reimbursement plan for the few expenses that won't go on the card.

The general feeling is that AmEx has better support for cardholders than their competitors, although I couldn't tell you how true that is.

I think you'd probably get a better answer if you could give an indication as to what was important to you. I use the AMEX Gold because the points can get me things I can use (Southwest Freq Flyer points, I live near BWI so Southwest is a good airline for me).

You could always try their new Plum card. The annual fee is $185 (waived in year one) but you can get a 2% discount if you pay your bill within 10 days of closing (if you spend over $5k, if you spend less you get 1%). I don't know how much extra cards are.

I've been happy with Chase, for a much smaller business. We get kickbacks of 1% to 3% on purchases, so if you're spending 400K a year, you should get at least $4,000 back. There may be a maximum, I'm not sure. If you really don't care about that much money, give it to your staff in gift cards.

They've got online card and account management, and decent support, at least in my experience.
